Vandetanib (Zactima)

Proposed brand name Zactima, Vandetamib is currently in clinical trials for approval by the United States Food and Drug Administration and is being developed by AstraZeneca. It is a proposed treatment for non-small-cell lung cancer (NSCLC) and is in clincial trials for mesothelioma treatment. The drug is a receptor antagonist for vascular endothelial growth factor receptors (VEGFR) and epidermal growth factor receptors (EGFR), and it operates by inhibiting the blood supply development to tumors and blocking the reception of specific chemicals by cells. The effect of those inhibitors causes cancerous cells to die and, ultimately, reduction in tumor size. Vandetanib is currently in Phase III development for treatment of non-small-cell lung cancer, but trials are continuing in Phase II studies to judge its effectiveness on glioma, thyroid, prostate, small-cell lung, and breast cancers.

Zactima is administered orally once a day and is being tested on patients with locally advanced or advanced non-small-cell lung cancer who have been unresponsive to other treatments. The trials conducted so far in Phases I, II, and III have been encouraging as patients who have not shown a response to previous treatments are showing response to Zactima. Current trials are comparing Zactima to other drugs to assess the compared survival rate, response duration, safety, and manageability of side effects among other factors.
