Predicting cancer-fighting abilities by computer
A new method of analyzing cellular activity based on computer modeling has predicted the anti-cancer properties of a multitude of molecules. The research, published in Molecular Cancer, outlines the functioning of ‘CoMet,' a computer tool that looks at the machinery of a cell and predicts which parts of the cell will have any effect on cancer.
John McDonald and Jeffery Skolnick worked together at the Georgia Institute of Technology to develop CoMet. Skolnick said, "This opens up the possibility of novel therapeutics for cancer and develops our understanding of why such metabolites work. CoMet provides a deeper understanding of the molecular mechanisms of cancer".
Metabolites, or small molecules that are produced by cells naturally in the daily running of their systems, can affect the expression of genes. Enzymes are protein chains that produce and destroy the metabolites in cells. According to Skolnick and McDonald, "By comparing the gene expression levels of cancer cells relative to normal cells and converting that information into the enzymes that produce metabolites, CoMet predicts metabolites that have lower concentrations in cancer relative to normal cells".
Their research showed that adding metabolites that are depleted by the abnormal cycles of cancer cells back to those cells will create anticancer properties. This is the basis for researchers' interest in kinase inhibitors. In the case of the study, leukemia cancer cells' growth was slowed by all nine metabolites that CoMet suggested. This treatment has many possible applications, and as Skolnick said, "While we have only performed cell proliferation assays, it is reasonable to speculate that some metabolites may also exhibit many other anticancer properties. These could be important steps on the road to a cure".
